VAKSIN DAN VAKSINASI IMUNOLOGI VETERINER
FAKULTAS KEDOKTERAN HEWAN UB 31 MARET 2020
AKTIF IMUNITAS
PASIF IMUNITAS Menerima antibodi
Membentuk antibodi
NATURAL
ARTIFICIAL
NATURAL
ARTIFICIAL
spesifik Limfosit B.
Mampu menginduksi long lived immune Menginduksi terbentuk sel memori Tidak memiliki efek samping
TIPE VAKSIN JARANG DIGUNAKAN LIVE VIRULENT VACCINES
A heterologous vaccine incorporates an organism that is antigenically related to the target infectious agent, but is adapted to another host species. The best example of such an approach is the use of human measles virus to protect against the related morbillivirus CDV. This procedure is no longer recommended.
TIPE VAKSIN YANG UMUM DIGUNAKAN
LIVE ATTENUATED VACCINES
This class of vaccine is the single most commonly used in veterinary medicine. These vaccines are based on the use of an intact and viable organism that has been ‘attenuated’ to reduce its virulence. Live attenuated organisms are capable of inducing lowlevel infection and replicating within the animal, but do not induce significant tissue pathology or clinical disease. Traditional means of attenuation might involve heating the vaccinal organisms, treating them with chemicals or growing them in unusual conditions (e.g.in particular cell lines or media, or by passaging them through an animal species to which they are not adapted). A more refined means of attenuation is to use molecular techniques to produce genetically modified organisms from which virulence genes have been deleted or modified. An example of this approach has been the development of vaccines for pseudorabies (Aujeszky’s disease) in swine, in which the thymidine kinase gene has been deleted from the causative herpesvirus.
KILLED VACCINE
A killed vaccine includes an organism that is antigenically intact, but clearly unable to replicate or induce pathology or clinical disease. There are various means of producing killed vaccines, most often by treating the vaccinal organisms with chemicals such as formalin, alcohol or alkylating agents.
VAKSIN SUBUNIT
A subunit vaccine does not contain an entire intact organism, but rather specific immunogenic structural proteins or metabolites derived from that organism. A good example of a subunit vaccine is the FeLV vaccine that incorporates glycoprotein 70 (gp70) extracted from the virus grown in cell culture. It is also possible to produce synthetic peptides based on knowledge of the structure of the antigenic protein of interest, but synthetic peptide vaccines require an adjuvant and produce immune responses of very restricted antigenic specificity.
MARKER VACCINES A further recent advance is the development of ‘marker vaccines’. Many infections are diagnosed by the detection of serum antibody as evidence of exposure (e.g. Lyme borreliosis, FIV), but as vaccination also induces serum antibody it has traditionally been difficult to discriminate between vaccinal and exposure titres. Marker vaccines have now been developed to make this distinction possible. An excellent example of such a product is the marker vaccine for infectious bovine rhinotracheitis (IBR). The virus contained in this product has deletion of the gene encoding surface glycoprotein E; therefore, if a cow has serum antibody to glycoprotein E, this must have been generated by exposure to field virus rather than by vaccination (269). Development of a marker vaccine requires parallel development of an appropriate diagnostic test.
KOMPONEN VAKSIN ANTIGEN
STABILIZER ADJUVANT ANTIBIOTIK PENGAWET
ADJUVANT Adjutants (Latin: adjuvare, to help)
are substances that when mixed with an antigen and injected with it, enhance the immunogenicity of the antigen.
Protect the antigen (vaccine) Non Antigenic material Used to boost the immune
response when:
The antigen has low immunogenicity
Only small amounts of antigen
Adjuvant: Elicit innate immune responses Increase costimulators signals Induce granuloma formation Increase cytokines, such as IL-12 for
stimulate growth & differentiation of immunocompetent cells Stimulate lymphocyte proliferation non
specifically
Postulated mode of action adjuvant
Prolongs antigen persistence
Enhance costimulatory signal
Induce granuloma formation
Stimulates lymphocytes nonspecifically
